A Novel Light-Responsive Gene BcCfaS Regulates the Photomorphogenesis and Virulence of Botrytis cinerea via Lipid Metabolism

摘要: Light is a fundamental environmental factor for living organisms on earth—not only as a primary energy source but also as an informational signal. In fungi, light can be used as an indicator for both time and space to control important physiological and morphological responses. Botrytis cinerea (B. cinerea) is a devastating phytopathogenic fungus that exploits light cues to optimize virulence and the balance between conidiation and sclerotia development, thereby improving its dispersal and survival in ecosystems. However, the components and mechanisms underlying these processes remain obscure. Here, we identify a novel light-signaling component in B. cinerea, BcCfaS, which encodes a putative cyclopropane fatty-acyl-phospholipid synthase. BcCfaS is strongly induced by light at the transcriptional level and plays a crucial role in regulating photomorphogenesis. Deletion of BcCfaS results in reduced vegetative growth, altered colony morphology, impaired sclerotial development, and enhanced conidiation in a light-dependent manner. Moreover, the mutant exhibits serious defects in stress response and virulence on the host. Based on a lipidomics analysis, a number of previously unknown fungal lipids and many BcCfaS-regulated lipids are identified in B. cinerea, including several novel phospholipids and fatty acids. Importantly, we find that BcCfaS controls conidiation and sclerotial development by positively regulating methyl jasmonate (MeJA) synthesis to activate the transcription of light-signaling components, revealing for the first time the metabolic base of photomorphogenesis in fungi. Thus, we propose that BcCfaS serves as an integration node for light and lipid metabolism, thereby providing a regulatory mechanism by which fungi adapt their development to a changing light environment. These new findings provide an important target for antifungal design to prevent and control fungal disease.

Effects of cotton straw amendment on soil fertility and microbial communities

摘要: Maintaining soil fertility, while controlling pollution from excessive chemical fertilizer application is important for keeping soil productivity of sustainable agriculture. Variety of straws have been used and proven to be good soil amendments for increasing soil organic matter (OM) and a range of additional soil nutrients. However, little is known about the utilization of cotton straw for soil amendment. To better understand the mechanism behind cotton straw soil amendments, investigations were performed upon cucumber seedlings, where changes to soil nutrients and microbial communities were investigated. The results revealed that the cotton straw application promoted the cucumber seedling growth by significantly increasing the soil OM, available nitrogen, available phosphorus, and available potassium. The concentration of cotton straw was positively correlated to both the number of the culturable microorganisms and also the total microbial biomass within soil. Furthermore, assessment of cotton straw application using Biolog metabolic profiling and phospholipid fatty acid analysis revealed that such application increased the microbial community metabolic activity, and markedly changed the structure of microbial community. 16S rRNA gene clone library construction and phylogenetic analysis of soil bacteria revealed - Proteobacteria sequences dominated the cotton straw amendment soil, comprising 27.8% of the total number of analyzed sequences, while they were less represented in control soil (13.4%). On the contrary, the Sphingobacteria (7.8%) and Verrucomicrobia (2.4%) in the cotton straw amendment soil decreased after application when compared to the control soil 15.2% and 15.2%.

Expression of inducible nitric oxide synthase in trophoblasts and deciduas in early medical abortion

摘要: The purpose of this study was to investigate the expression of inducible nitric oxide synthase in trophoblasts and deciduas in early medical abortion, and study the relationship of medical abortion through mifepristone and inducible nitric oxide synthase (iNOS) in early pregnancy. Expression of iNOS in trophoblasts and deciduas was detected by both hybridization and immunohistochemical assay in 40 patients (experimental group); the positive expression of iNOS was represented by number density (N/S) and positive unit (Pu) using computer color magic image analysis system (CMIAS). All results were compared with that obtained from vacuum aspiration. In the experimental group, N/S and Pu in trophoblasts were 0.120 ± 0.010 and 15.3 ± 2.6, respectively, while in the control group, they were 0.021 ± 0.003 and 3.1 ± 0.5, respectively, and there were significant differences between the two groups. By immunohistochemical assay, N/S and Pu were 0.090 ± 0.010, 10.24 ± 1.55 0.016 ± 0.002, 1.26 ± 0.33 in the trophoblasts of the two groups; there were also significant differences between the two groups. There were lower iNOS expressions in deciduas by in situ hybridization and immunohistochemical assay, and the difference between the two groups was not significant.It was concluded that mifepristone induced medical abortion through the expression of iNOS in trophoblasts but not in deciduas.

miRNAs in non-alcoholic fatty liver disease

摘要:

Non-alcoholic fatty liver disease (NAFLD) is a major cause of liver cirrhosis and hepatocellular carcinoma and is a considerable threat to public health. miRNAs are important post-transcriptional regulators of gene expression, and the dysregulation of miRNAs is involved in various biological processes in the liver, including lipid homeostasis, inflammation, apoptosis, and cell proliferation. Recently, a number of studies have described the association between miRNAs and NAFLD progression and have shown that circulating miRNAs reflect histological changes in the liver. Therefore, circulating miRNAs have potential use for the evaluation of NAFLD severity. In this review, we discuss the involvement of miRNAs in NAFLD pathogenesis and the key role of miRNAs in the screening, diagnosis, and staging of NAFLD.

Nonalcoholic fatty liver disease (NAFLD) covers a spectrum of liver disorders ranging from simple steatosis to advanced pathologies, including nonalcoholic steatohepatitis and cirrhosis. NAFLD significantly contributes to morbidity and mortality in developed societies. Insulin resistance associated with central obesity is the major cause of hepatic steatosis, which is characterized by excessive accumulation of triglyceride-rich lipid droplets in the liver. Accumulating evidence supports that dysregulation of adipose lipolysis and liver de novo lipogenesis (DNL) plays a key role in driving hepatic steatosis. In this work, we reviewed the molecular mechanisms responsible for enhanced adipose lipolysis and increased hepatic DNL that lead to hepatic lipid accumulation in the context of obesity. Delineation of these mechanisms holds promise for developing novel avenues against NAFLD.

Relationship of endothelial nitric oxide synthase gene polymorphism with blood pressure, lipid profile

摘要: To study the relationship of the polymorphism of endothelial nitric oxide synthase (eNOS) gene and blood pressure, lipid profiles and blood glucose level. By using PCR-RFLP, the eNOS Glu298Asp gene polymorphism was detected in 184 patients with essential hypertension and 196 matched healthy individuals with normal blood pressure. Taking into account eNOS Glu298Asp polymorphisms, the relationship of blood pressure with triglycerides (TG), total cholesterol (TC), high density lipoprotein (HDL), low density lipoprotein (LDL) and blood glucose level was analyzed. The distribution of eNOS Glu298Asp polymorphism had no significant difference between different blood pressure groups and gender groups, but there was a significant difference between different age groups, diastolic blood pressure groups or BMI groups ( < 0.05). Asp/Asp genotype significantly increased the risk of hypertension in individuals with serum TC above 5.4 mmol/L ( = 0.03, = 2.65). eNOS Glu298Asp polymorphism and serum lipid could synergistically modulate the blood pressure. eNOS Asp/Asp genotype could significantly increase the risk of hypertension in individuals with serum TC over 5.4 mmol/L. eNOS Glu298Asp in combination with serum TC could be used to predict the risk of hypertension.

New definition of metabolic dysfunction-associated fatty liver disease with elevated brachial-ankle pulse

摘要: A new definition of metabolic dysfunction-associated fatty liver disease (MAFLD) has recently been proposed. We aim to examine the associations of MAFLD, particularly its discordance from non-alcoholic fatty liver disease (NAFLD), with the progression of elevated brachial-ankle pulse wave velocity (baPWV) and albuminuria in a community-based study sample in Shanghai, China. After 4.3 years of follow-up, 778 participants developed elevated baPWV and 499 developed albuminuria. In comparison with the non-MAFLD group, the multivariable adjusted odds ratio (OR) of MAFLD group for new-onset elevated baPWV was 1.25 (95% confidence interval (CI) 1.01–1.55) and 1.35 (95% CI 1.07–1.70) for albuminuria. Participants without NAFLD but diagnosed according to MAFLD definition were associated with higher risk of incident albuminuria (OR 1.77; 95% CI 1.07–2.94). Patients with MAFLD with high value of hepamet fibrosis score or poor-controlled diabetes had higher risk of elevated baPWV or albuminuria. In conclusion, MAFLD was associated with new-onset elevated baPWV and albuminuria independently of body mass index, waist circumference, and hip circumference. Individuals without NAFLD but diagnosed as MAFLD had high risk of albuminuria, supporting that MAFLD criteria would be practical for the evaluation of long-term risk of subclinical atherosclerosis among fatty liver patients.

Responses of bacterial strains isolated from drinking water environments to N-acyl-L-homoserine lactones

摘要: Often as a result of biofilm formation, drinking water distribution systems (DWDS) are regularly faced with the problem of microbial contamination. Quorum sensing (QS) systems play a marked role in the regulation of microbial biofilm formation; thus, inhibition of QS systems may provide a promising approach to biofilm formation control in DWDS. In the present study, 22 bacterial strains were isolated from drinking water-related environments. The following properties of the strains were investigated: bacterial biofilm formation capacity, QS signal molecule N-acyl-L-homoserine lactones (AHLs) production ability, and responses to AHLs and AHL analogs, 3-chloro-4-(dichloromethyl)-5-hydroxy-2(5H)-furanone (MX) and 2(5H)-furanone. Four AHLs were added to developed biofilms at dosages ranging from 0.1?nmol·L to 100 nmol·L . As a result, the biofilm growth of more than 1/4 of the isolates, which included AHL producers and non-producers, were significantly promoted. Further, the biofilm biomasses were closely associated with respective AHLs concentrations. These results provided evidence to support the idea that AHLs play a definitive role in biofilm formation in many of the studied bacteria. Meanwhile, two AHLs analogs demonstrated unexpectedly minimal negative effects on biofilm formation. This suggested that, in order to find an applicable QS inhibition approach for biofilm control in DWDS, the testing and analysis of more analogs is needed.

Optimization of pretreatment of Jatropha oil with high free fatty acids for biodiesel production

摘要: A central composite rotatable design and response surface methodology were used in order to investigate the individual and combined effects of the ethanol-to-oil ratio, H SO concentration, temperature and time of reaction on the reduction of free fatty acid (FFA) in jatropha oil. A quadratic polynomial model relating the reaction variables with FFA reduction was developed, presenting a good coefficient of determination ( = 0.893). For reducing FFA to less than 1%, the optimal combination was found to be 0.62 v·v ethanol-to-oil ratio (14.9 v·v ethanol-to-FFA ratio), 1.7% v·v H SO concentration, and 79 min reaction time at a reaction temperature of 54°C. These results are of great relevance to maximize methyl esters formation by transesterification using an alkaline catalyst.

Role of Akkermansia muciniphila in the development of nonalcoholic fatty liver disease: current knowledge

摘要: Nonalcoholic fatty liver disease (NAFLD) is a hepatic manifestation of metabolic syndrome and a common cause of liver cirrhosis and cancer. Akkermansia muciniphila (A. muciniphila) is a next-generation probiotic that has been reported to improve metabolic disorders. Emerging evidence indicates the therapeutic potential of A. muciniphila for NAFLD, especially in the inflammatory stage, nonalcoholic steatohepatitis. Here, the current knowledge on the role of A. muciniphila in the progression of NAFLD was summarized. A. muciniphila abundancy is decreased in animals and humans with NAFLD. The recovery of A. muciniphila presented benefits in preventing hepatic fat accumulation and inflammation in NAFLD. The details of how microbes regulate hepatic immunity and lipid accumulation in NAFLD were further discussed. The modulation mechanisms by which A. muciniphila acts to improve hepatic inflammation are mainly attributed to the alleviation of inflammatory cytokines and LPS signals and the downregulation of microbiota-related innate immune cells (such as macrophages). This review provides insights into the roles of A. muciniphila in NAFLD, thereby providing a blueprint to facilitate clinical therapeutic applications.

Chinese herbal formula, improves insulin resistance in overweight/obese subjects with non-alcoholic fatty

摘要: Lingguizhugan Decoction (LGZG) has been investigated in basic studies, with satisfactory effects on insulin resistance in non-alcoholic fatty liver disease (NAFLD). This translational approach aimed to explore the effect and underlying mechanism of LGZG in clinical setting. A randomized, double-blinded, placebo-controlled trial was performed. A total of 243 eligible participants with NAFLD were equally allocated to receive LGZG (two groups: standard dose and low dose) or placebo for 12 weeks on the basis of lifestyle modifications. The primary efficacy variable was homeostasis model assessment of insulin resistance (HOMA-IR). Analyses were performed in two populations in accordance with body mass index (BMI; overweight/obese, BMI ≥ 24 kg/m2; lean, BMI < 24 kg/m 2). For overweight/obese participants, low-dose LGZG significantly decreased their HOMA-IR level compared with placebo (−0.19 (1.47) versus 0.08 (1.99),P = 0.038). For lean subjects, neither dose of LGZG showed a superior effect compared with placebo. Methylated DNA immunoprecipitation sequencing and real-time qPCR found that the DNA N6-methyladenine modification levels of protein phosphatase 1 regulatory subunit 3A (PPP1R3A) and autophagy related 3 (ATG3) significantly increased after LGZG intervention in overweight/obese population. Low-dose LGZG effectively improved insulin resistance in overweight/obese subjects with NAFLD. The underlying mechanism may be related to the regulation of DNA N6-methyladenine modification of PPP1R3A and ATG3. Lean subjects may not be a targeted population for LGZG.

Type 2 diabetic patients with non-alcoholic fatty liver disease exhibit significant haemorheological

摘要: Haemorheological abnormalities have been described in diabetes mellitus, as well as in non-alcoholic fatty liver disease (NAFLD). However, the relationship between the changes in liver fat content and haemorheology is unknown. The current study aims to show the correlation between haemorheological parameters and intrahepatic lipid content (IHLC) in patients with type 2 diabetes. The serum biochemical markers, such as fasting plasma glucose (FPG), haemoglobin A1c (HbA1c), liver enzymes, lipid profiles, and haemorheological properties, were examined. IHLC was quantified using proton magnetic resonance spectroscopy ( H-MRS) scanning of the liver. A significant correlation was observed between IHLC and whole blood viscosity at high, middle, and low shear rates. IHLC also positively correlated with haematocrit, the reduced whole blood viscosity at low and middle shear rates, and the erythrocyte aggregation index. Diabetic patients with NAFLD exhibited significant haemorheological abnormalities compared with patients without NAFLD. In summary, haemorheological disorders are linked to non-alcoholic fatty liver in type 2 diabetes.

Co-fermentation of waste activated sludge with food waste for short-chain fatty acids production: effect

摘要: Effect of pH ranging from 4.0 to 11.0 on co-fermentation of waste activated sludge (WAS) with food waste for short-chain fatty acids (SCFAs) production at ambient temperature was investigated in this study. Experimental results showed that the addition of food waste significantly improved the performance of WAS fermentation system, which resulted in the increases of SCFAs production and substrate reduction. The SCFAs production at pH 6.0, 7.0, 8.0, and 9.0 and fermentation time of 4 d was respectively 5022.7, 6540.5, 8236.6, and 7911.7 mg COD·L , whereas in the blank tests (no pH adjustment, pH 8.0 (blank test 1), no food waste addition, pH 8.0 (blank test 2), and no WAS addition (blank test 3)) it was only 1006.9, 971.1, and 1468.5 mg COD·L , respectively. The composition of SCFAs at pH from 6.0 to 9.0 was also different from other conditions and propionic acid was the most prevalent SCFA, which was followed by acetic and n-butyric acids, while acetic acid was the top product under other conditions. At pH 8.0 a higher volatile suspended solids (VSS) reduction of 16.6% for the mixture of WAS and food waste than the sole WAS indicated a synergistic effect existing in fermentation system with WAS and food waste. The influence of pH on the variations of nutrient content was also studied during anaerobic fermentation of the mixture of WAS and food waste at different pH conditions. The release of increased with fermentation time at all pH values investigated except 4.0, 5.0 and in blank test one. The concentrations of soluble phosphorus at acidic pHs and in the blank test one were higher than those obtained at alkaline pHs. Ammonia and phosphorus need to be removed before the SCFAs-enriched fermentation liquid from WAS and food waste was used as the carbon source.

Analysis of Brucella cellular fatty acids

摘要: To acquire data of Brucella cellular fatty acids (CFAs) and probe into the possibility of utilizing CFAs information in typing Brucella, 19 reference strains were subjected to CFAs study. After all strains were inoculated on Brucella Agar plates, the cells were harvested, saponificated, methylated and extracted to provide fatty acid methylesters for gas chromatography (GC) analysis. Based on the CFAs data matrix, a dendrogram of 19 reference strains was generated by SPSS11.5 software package. The results showed that 19 reference strains were divided into five clusters: cluster 1 included (bv. 1, 2, 3, 5) and ; cluster 2 included (bv. 3, 4, 5, 6) and (bv. 1, 2, 3); cluster 3 included (bv. 1, 2, 7, 9) and ; cluster 4 was (bv. 4); and cluster 5 was . Typing Brucella by GC analysis of CFAs is a good method to reflect drug resistance of Brucella, and the classification is beneficial for clinical therapy. It also provides a new result of typing and demonstrates that the traditional classification is not completely reasonable. CFAs analysis may identify (bv. 4) and .
